Genetic diversity analysis of sugarcane genotypes by SSR markers

Abstract

Sugarcane is a potential sugar producing crop in Bangladesh and has scope to increase this potentiality through genetic improvement. The investigation was conducted for genetic diversity analysis of ten sugarcane genotypes (Isd 36, Isd 37, Isd 38, Isd 39, Isd 40, BSRI Akh 41, Bombay, Babulal, Q 83 and Misrimala) using SSR markers. BSRI Akh 41, Bombay, Babulal, Q 83 and Misrimala were chewing type of sugarcane. Two SSR primers SMC336BUQ and SMC334BS were selected based on initial screening of five primers (SMC36BUQ, SMC119CG, SMC319CG, SMC334BS and SMC336BS). Two microsatellite primer pairs were amplified a total number of 16 bands from the ten genotypes and visualized through 2% Agarose gel electrophoresis. The sizes of the amplified bands ranged from 80 to 184bp. Genetic diversity or polymorphic information content (PIC) per primer pair was 0.80 for all loci across the ten cultivars evaluated. Both the primer pairs were the most polymorphic marker for ten genotypes with PIC values of 0.80. The number of genotypes with unique banding patterns distinguished by the two SSR markers was 100%. The highest linkage distance was recorded between the varieties Isd 36 with Isd 37, Isd 39 and Isd 40, respectively. Genetic diversity analysis indicated that genotypes developed later showed more genetic distances than earlier developed genotypes as well as local chewing genotypes maintained more genetic distances than bred chewing genotypes. The results of the present investigation revealed that two SSR markers were able to identify genetic differences among the sugarcane genotypes.
